How to make an unnatural bioweapon?

Imagine you can produce a virus with these 4 new nucleotides, adding S, B, P and Z ... to the usual A, C, G, T?  What if they are only partially viable in an infected cell (in terms of the virus attempting to reproduce itself using stolen chemical machinery)?  Wouldn't this be useful in poisoning the victim in a unique way, that is hard to detect if you are only looking for A, C, G and T?

Envisioned this way back in 1974, but I didn't see any way to do it.  Was thinking of A, C, G and T with carbon replaced by silicon.  But apparently there are more chemically compatible alternatives.
